RESULTS Ex vivo studied the effect of stimulation of α1-adrenergic receptors by methoxamine on heart rate and systolic pressure of the left ventricle of rat heart. Stimulation of α1-AR methoxamine at a concentration of 10-10 M to 3 minutes resulted in slowing of the heart (12%) and reducing the force of contraction of 12%. After 5 minutes, the heart rate decreased by 28.7% (p≤0.01), for 15 minutes, 35% (p≤0.05). Throughout the experiment the maximum decrease in contractility of the left ventricle was 27,8% (p≤0.05). Immediately after the introduction methoxamine at a concentration of 10-8 M, an increase in the contractility of the left ventricle by 12% (p≤0.05), the value of LVP changed with 62,35±5,04 to 69,8±6.8 mm Hg. On the 7minute perfusion was observed a decrease in contraction force by 65% (p≤0.01). Methoxamine at a concentration of 10-8 M caused a reduction in heart rate. The maximum reduction in heart rate was 29.5% (p≤0.05). Thus, stimulation of α1-AR in Langendorff perfused heart was led to bradycardia regardless of the concentration of agonist. Methoxamine at a concentration of 10-8 M resulted in mixed effects contractility. Experiments with different concentrations of the nonselective agonist showed a positive chronoinotropic dependence.
